[212Pb]Pb-PSV359 is a peptide-based radiopharmaceutical designed to deliver the alpha-emitting radionuclide lead-212 (212Pb) specifically to tumor sites expressing fibroblast activation protein alpha (FAP, also known as FAP-α). FAP is highly expressed on cancer-associated fibroblasts in the tumor microenvironment of many solid tumors. The drug consists of a cyclic peptide ligand (PSV359), discovered and optimized via phage display for high affinity and specificity to human FAP, conjugated with 212Pb. Upon intravenous administration, [212Pb]Pb-PSV359 binds selectively to FAP-expressing cells, delivering targeted alpha-particle radiation that induces cytotoxicity in both tumor cells and supportive stromal elements. Preclinical studies have demonstrated strong anti-tumor efficacy with minimal off-target toxicity. The agent is being developed by Perspective Therapeutics for use as a targeted therapy in patients with advanced solid tumors that are positive for FAP expression[1][3][5][6][7].
